Stability of Cholesterol; Cholesterol-5β,6β-Epoxide; Cholesta-3,5-Dien-7-One, and 5α-Cholestane in Solution During Short-Term Storage

چکیده
Because cholesterol and its oxide products (COPs) are used as standards during analysis, their stability and autoxidation were assessed under typical storage conditions. Individual toluene solutions of cholesterol; cholesterol5β, 6β-epoxide; cholesta-3,5-dien-7-one; and 5α-cholestane were prepared and aliquots were stored at 23 ± 2°C, 4°C, and -20°C for 7 days. Samples were removed from storage, dried under nitrogen, derivatized and analyzed using gas chromatography-flame ionization detection and gas chromatography-mass spectrometry. β-sistosterol and 5-colest-ene-7-one were found to be a suitable recovery internal standard and an instrument internal standard, respectively. All compounds in solution had no detectable artifact generation during 7 days of storage; thus, they were stable. Storage temperatures of 23 ± 2°C, 4°C, and -20°C did not affect stability. Chromatograms revealed contaminants and other peaks likely associated with incomplete derivatization of some compounds; potential causes for the unusual lack of derivitization are discussed.

In the present study, we show that four unusual cholesterol precursors, 5α-cholesta-8,14-dien-3β-ol, 4-methyl-5α-cholesta-8,24-dien-3β-ol (4-methylzymosterol), 4,4,14-trimethyl-5α-cholesta-8,24-dien-3β-ol (lanosterol), and 4,4-dimethyl-5α-cholesta-8,24-dien-3β-ol (4,4-dimethylzymosterol) are present in testes of the limpet Cellana grata, which is part of one of the most primitive gastropod fami...
متن کاملUnravelling new pathways of sterol metabolism: lessons learned from in-born errors and cancer
PURPOSE OF REVIEW To update researchers of recently discovered metabolites of cholesterol and of its precursors and to suggest relevant metabolic pathways. RECENT FINDINGS Patients suffering from inborn errors of sterol biosynthesis, transport and metabolism display unusual metabolic pathways, which may be major routes in the diseased state but minor in the healthy individual. Although quanti...
متن کاملCholesterol epoxide hydrolase and cancer.
Cholesterol epoxide hydrolase (ChEH) catalyzes the hydration of cholesterol-5,6-epoxides (5,6-EC) into cholestane-3β,5α,6β-triol. ChEH is a hetero-oligomeric complex called the anti-estrogen binding site (AEBS) comprising 3β-hydroxysterol-Δ(8)-Δ(7)-isomerase (D8D7I) and 3β-hydroxysterol-Δ(7)-reductase (DHCR7). D8D7I and DHCR7 regulate cholesterol biosynthesis, fetal development and growth, tumo...
متن کاملEffects of Various Cooking and Re-heating Methods on Cholesterol Oxidation Products of Beef Loin
The objective of this study was to define the effects of various cooking and re-heating methods (pan roasting, steaming, oven grilling and micro-waving) on the cholesterol and formation of cholesterol oxidation products in beef loin during storage at 4°C. Raw samples showed lower total cholesterol content than cooked products sampled during storage for 6 d. The following cholesterol oxidation p...
متن کاملOxysterols from human bile induce apoptosis of canine gallbladder epithelial cells in monolayer culture.
Oxysterols have been detected in various mammalian organs and blood. Biliary epithelium is exposed to high concentrations of cholesterol, and we have identified three keto-oxysterols (cholest-4-en-3-one, cholesta-4,6-dien-3-one, cholesta-3,5-dien-7-one) in human bile and gallstones. Because the effects of oxysterols on biliary physiology are not well defined, we investigated their biological ef...
